AOD-9604: The Fat Loss Fragment Peptide

What Is AOD-9604?
AOD-9604 (Anti-Obesity Drug 9604) is a synthetic peptide fragment corresponding to amino acids 176-191 of human growth hormone, with a tyrosine residue added at the beginning. It was developed by Professor Frank Ng at Monash University in Melbourne, Australia, with the goal of isolating the lipolytic (fat-burning) activity of growth hormone from its growth-promoting and blood sugar-raising effects.
Growth hormone is well-known for its ability to reduce body fat, but full-length GH also promotes growth of muscles and organs, raises blood sugar, and can cause joint pain and edema. AOD-9604 was designed to capture just the fat metabolism benefits without these other effects.
Mechanism of Action
AOD-9604 works through the same pathways that the C-terminal fragment of growth hormone uses to regulate fat metabolism:
- Stimulates lipolysis -- AOD-9604 activates beta-3 adrenergic receptors on adipocytes (fat cells), triggering the breakdown of stored triglycerides into free fatty acids and glycerol. These fatty acids are then available for energy production.
- Inhibits lipogenesis -- The peptide reduces the rate at which new fat is formed from circulating nutrients, particularly in the presence of excess caloric intake.
- No effect on blood sugar -- Unlike full-length GH, AOD-9604 does not interfere with insulin sensitivity or glucose metabolism. Clinical trials confirmed no changes in fasting glucose, insulin, or HbA1c.
- No effect on IGF-1 -- AOD-9604 does not stimulate IGF-1 production, which means it lacks the muscle-building and organ-growth effects of GH.
Clinical Trial Results
AOD-9604 progressed to phase IIb clinical trials in obese humans, making it one of the few peptides with substantial human data:
In a 12-week randomized, double-blind, placebo-controlled trial of 300 obese participants, the group receiving 1 mg oral AOD-9604 daily lost significantly more weight than placebo. The study demonstrated a favorable safety profile with no serious adverse events, no changes in IGF-1, glucose tolerance, or lipid profiles.
While the oral trial results were statistically significant, the magnitude of weight loss was modest compared to modern GLP-1 agonists. Injectable forms are thought to be more potent due to higher bioavailability, and research interest has shifted toward subcutaneous administration protocols.
Dosing Protocol
- Standard dose: 300 mcg subcutaneous injection daily
- Timing: Morning, on an empty stomach (fasting enhances fat mobilization)
- Injection site: Abdominal subcutaneous tissue (near the target fat deposits)
- Cycle length: 12-20 weeks
Peptide Calculator Setup
AOD-9604 commonly comes in 5 mg vials. Using the Peptide Calculator Plus:
- 5 mg vial + 1 mL BAC water = 5,000 mcg/mL
- For 300 mcg: draw 6 units on a U-100 syringe
- One vial lasts approximately 16-17 days at 300 mcg/day
AOD-9604 vs Other Fat Loss Peptides
How does AOD-9604 compare to other fat loss peptides?
- vs Semaglutide/Tirzepatide -- GLP-1 agonists produce dramatically greater weight loss (15-22% body weight). AOD-9604's advantage is its favorable side effect profile (no GI issues) and lack of hormonal disruption.
- vs CJC-1295/Ipamorelin -- GH secretagogues increase overall GH, which burns fat but also has muscle-building and other effects. AOD-9604 is more targeted to fat loss specifically.
- vs HGH Fragment 176-191 -- These are essentially the same peptide. AOD-9604 includes a tyrosine modification that may improve stability.
Safety and Side Effects
AOD-9604 demonstrated an excellent safety profile in clinical trials. No serious adverse events were attributed to the peptide. Reported effects include mild injection site reactions, occasional headache, and mild gastrointestinal discomfort. The TGA (Therapeutic Goods Administration) of Australia has even approved AOD-9604 as a food supplement ingredient, indicating its recognized safety at oral doses.
Practical Considerations
AOD-9604 works best as part of a comprehensive approach that includes caloric control and exercise. It is not a magic bullet for weight loss but may enhance fat mobilization when combined with a proper diet. The lack of muscle-building effects means it does not alter body composition in the same way as GH, so it is most appropriate for individuals specifically targeting fat loss rather than overall body recomposition.
